DSIP

DSIP (delta sleep-inducing peptide) is a nonapeptide (nine amino acids) first isolated from rabbit brain tissue in the 1970s during research into the neurochemical basis of sleep. It is one of the earliest-discovered “sleep peptides” and has a research history spanning several decades, though the literature is comparatively smaller and older than for many other peptides.

What Is DSIP?

DSIP was originally identified by researchers studying blood drawn from rabbits during electrically induced delta-wave sleep, with the isolated peptide named for its apparent association with that sleep stage. Its proposed mechanisms, as described in the older literature, include:

  • Neuromodulatory activity — DSIP has been studied for potential interactions with GABAergic and other neurotransmitter systems involved in sleep-wake regulation, though a definitive receptor has not been firmly established in the literature.

It’s worth noting that DSIP’s name reflects the hypothesis under which it was originally studied rather than a fully confirmed, mechanistically established function — a distinction the original researchers and subsequent reviewers have themselves noted, since later studies produced inconsistent results regarding its actual effect on delta-wave sleep.

DSIP’s research literature is older and more limited in volume compared to many peptides currently studied, with most primary research published between the 1970s and 1990s:
